Initiation of the Meiotic Prophase-to-metaphase Transition in Mammalian Oocytes

چکیده
Mammalian oocytes are arrested in meiotic prophase for prolonged periods, and then resume meiosis in response to luteinizing hormone from the pituitary, which binds to the somatic cells surrounding the oocyte. This chapter reviews current knowledge and questions about the molecular mechanisms that mediate communication between the somatic cells and oocyte to control the prophase-to-metaphase transition. Zinc maintains prophase I arrest in mouse oocytes through regulation of the MOS-MAPK pathway.
Meiosis in mammalian females is marked by two arrest points, at prophase I and metaphase II, which must be tightly regulated in order to produce a haploid gamete at the time of fertilization. The transition metal zinc has emerged as a necessary and dynamic regulator of the establishment, maintenance, and exit from metaphase II arrest, but the roles of zinc during prophase I arrest are largely u...
متن کاملMeiotic maturation of mouse oocytes in vitro: protein synthesis in nucleate and anucleate oocyte fragments.
Nucleate and anucleate fragments of mouse oocytes have been isolated following treatment of fully grown oocytes with cytochalasin B. The nucleate oocyte fragments resume meiosis in vitro, progressing from dictyate of the first meiotic prophase to metaphase II ('meiotic maturation'), and exhibit all of the changes in protein synthesis normally associated with meiotic maturation of mouse oocytes....
متن کاملChromatin Spread Preparations for the Analysis of Mouse Oocyte Progression from Prophase to Metaphase II.
Chromatin spread techniques have been widely used to assess the dynamic localization of various proteins during gametogenesis, particularly for spermatogenesis. These techniques allow for visualization of protein and DNA localization patterns during meiotic events such as homologous chromosome pairing, synapsis and DNA repair. While a few protocols have been described in the literature, general...
متن کاملProphase I arrest and progression to metaphase I in mouse oocytes are controlled by Emi1-dependent regulation of APCCdh1
Mammalian oocytes are arrested in prophase of the first meiotic division. Progression into the first meiotic division is driven by an increase in the activity of maturation-promoting factor (MPF). In mouse oocytes, we find that early mitotic inhibitor 1 (Emi1), an inhibitor of the anaphase-promoting complex (APC) that is responsible for cyclin B destruction and inactivation of MPF, is present a...
متن کاملComparative analysis of meiotic progression in female mice bearing mutations in genes of the DNA mismatch repair pathway.
The DNA mismatch repair (MMR) family functions in a variety of contexts to preserve genome integrity in most eukaryotes. In particular, members of the MMR family are involved in the process of meiotic recombination in germ cells. MMR gene mutations in mice result in meiotic disruption during prophase I, but the extent of this disruption often differs between male and female meiocytes. To addres...
